Abstract
This dataset contains electrochemical data obtained from ECL chronoamperometry experiments using magnetic bead–antibody–[Ru(bpy)₃]²⁺ conjugates at varying concentrations, spotting five different concentrations (0, 0.033, 0.1, 0.33 and 1 nM) per rectangular-shaped screen-printed electrode. The dataset aims to quantify the luminescent response of the system and support the image-based quantitative analytical methods of the platform.

The experimental conditions of the chronoamperometry experiments were:
	- Analyte: Magnetic bead-Ab-[Ru(bpy)₃]²⁺ at concentrations of 0, 0.033, 0.1, 0.33 or 1 nM
	- Coreactant/Electrolyte: 180 mM tripropylamine (TPrA) in 0.3 M phosphate buffer, pH 6.8
	- Electrode Configuration (Screen-printed electrodes):
		- Working electrode: GST4500 C
		- Reference electrode: EDAG 725A Ag
		- Counter electrode: GST4500 C
	- Chronoamperometric Protocol:
		- Frameroi and background images (bg): No electrochemical signal applied; images used for ROI selection and background reference.
 		- ECL recordings: 
			- Open circuit potential (OCP) for 2 seconds 
			- Chronoamperometry (CA) step to 2 V vs Ag for 5 seconds 
	- Temperature = room temperature
	- Recording parameters:
		Low-light OV9281 monochrome camera settings: analogue gain 15×, duration 8 s, framerate 1/8 frames/s, resolution 1280×800 pixels, brightness 0, contrast 1, noise reduction off, sensor mode 1280×800-Y10_1X10/RAW with YUV420 preview, buffer count = 10.
Note: For frameroi images, all settings except resolution were set to automatic; resolution fixed at 1280×800 pixels.
	- Processing done for visualisation purposes (only applied to processed frames) = automatic brightness and contrast adjustment.

Content of the files 
• “C1_2V_png.png” – “C5_2V_png.png” contains PNG image frames of the ECL signal recorded during OCP-CA experiments.
• “C1_2V_png_processed.png” – “C5_2V_png_processed.png” contains processed PNG image frames of the ECL signal recorded during OCP–CA experiments.
• “C1_2V_tiff.tiff” – “C5_2V_tiff.tiff” contains TIFF image frames of the ECL signal recorded during OCP–CA experiments.
• “C1_bg_png.png” – “C5_bg_png.png” contains PNG background images acquired.
• “C1_bg_tiff.tiff” – “C5_bg_tiff.tiff” contains TIFF background images acquired.
• “C1_frameroi.png” – “C5_frameroi.png” contains PNG image frames of the initial electrode image used for ROI selection during image analysis.
• "Calibration_multiplex_SAP.csv" contains ECL intensity data extracted from the selected ROIs of the image frames.
• "OCP-2s_CA_2V-5s_electrochemical_data_multiplex_SAP.csv" contains electrochemical data of the OCP-CA experiments.

File Specifics
• 30 captured image frames
The dataset includes image files in PNG and TIFF formats captured and saved using custom-developed platform software.	
• 2 Excel files
The dataset includes two data files provided in CSV (Comma-Separated Values) format, containing the numerical outputs generated by the platform. The files were combined using Microsoft Excel for Microsoft 365 (MSO Version 2111, Build 16.0.19426.20260) and exported as CSV files.

Filename Convention
• 90 captured image frames
Each filename follows the patterns:
[SampleID]_frameroi.[FileExtension]; [SampleID]_[Voltage]_[FileType].[FileExtension]; [SampleID]_[Voltage]_[FileType]_processed.[FileExtension]; [SampleID]_bg_[FileType].[FileExtension]; 

Where:
	- SampleID: Sequential identifier of the measurement type C (multiplex magnetic bead-Ab-[Ru(bpy)₃]²⁺ experiments).
	- frameroi: Indicates a frame used for ROI selection during analysis.
	- Voltage: Voltage applied during the chronoamperometric measurement (only when applied).
	- processed: Indicates that the frame has been processed by the software.
	- FileType/FileExtension: PNG or TIFF.
	- bg: Indicates a background image.

Codebook of Variables
• 2 Excel files
The dataset includes two tabular quantitative files in CSV (.csv) format. The columns in each file represent the following variables:

1. "OCP-2s_CA_2V-5s_electrochemical_data_multiplex-SAP.csv":
• Time (s) = Time elapsed from the start of the electrochemical measurement, in seconds (s).
• Measured Potential (V) = Potential response measured during open-circuit potential (OCP), in volts (V), for each experiment.
• Measured Current (A) = Electrical current recorded in response to the applied potential during chronoamperometry (CA), in amperes (A), for each experiment.

2. "Calibration_multiplex_SAP.csv":
• Experiment = Identifier of the experiment number.	
• [Ab-Ru] (nM) = Nominal concentration of magnetic bead-Ab-[Ru(bpy)₃]²⁺, in nanomolar (nM) 	
• ECL_png_raw (counts) = Raw ECL intensity measured from the selected ROIs in PNG image frames, in counts.
• ECL_png_raw-bg (counts) = Background-subtracted ECL intensity measured from the selected ROIs in PNG image frames, in counts.
